研究領(lǐng)域
真核細(xì)胞發(fā)育和分化過(guò)程中，功能基因組的表達(dá)受到精細(xì)而廣泛的調(diào)控，主要通過(guò)相應(yīng)的調(diào)控元件如啟動(dòng)子增強(qiáng)子等實(shí)現(xiàn)。近年來(lái)的研究發(fā)現(xiàn)，真核細(xì)胞內(nèi)染色體組蛋白的特定修飾與基因調(diào)控元件密切相關(guān)，如H3K4me3特異性標(biāo)記啟動(dòng)子，H3K4me特異性標(biāo)記增強(qiáng)子，而H3K27ac標(biāo)記活化的調(diào)控原件。通過(guò)組蛋白特定修飾相對(duì)應(yīng)的抗體進(jìn)行高通量的Chip-seq實(shí)驗(yàn)，在全基因組范圍內(nèi)揭示功能基因組的調(diào)控元件，并結(jié)合生物信息學(xué)的方法統(tǒng)計(jì)分析與調(diào)控元件相結(jié)合的轉(zhuǎn)錄因子，從而確定真核細(xì)胞發(fā)育和分化的關(guān)鍵基因及其調(diào)控網(wǎng)絡(luò)。系統(tǒng)性的研究生命過(guò)程中功能基因組表達(dá)和相互之間的調(diào)控關(guān)系，從而繪制基因調(diào)控回路，全面解析生命過(guò)程。
