d.生物順磁核磁:利用稀土順磁效應(yīng)揭示生物大分子的作用機(jī)制和作用構(gòu)象變化、小分子與大分子作用的高分辨結(jié)構(gòu);有效藥物先導(dǎo)分子的篩選與優(yōu)化;蛋白質(zhì)動(dòng)態(tài)學(xué)研究的新方法與新技術(shù)。
e. 蛋白質(zhì)無(wú)細(xì)胞合成:具有重要功能蛋白質(zhì)的無(wú)細(xì)胞合成探索,其中主要包括膜蛋白的體外合成與標(biāo)記
核磁共振研究中蛋白質(zhì)標(biāo)記方法大多集中于巰基化學(xué)反應(yīng),這是由于L-半胱氨酸的豐度較低并且?guī)基的化學(xué)選擇性高的緣故。在蛋白質(zhì)順磁標(biāo)記領(lǐng)域方面,我們提出了利用末端烯烴與巰基的Michael加成反應(yīng)標(biāo)記蛋白質(zhì),這類反應(yīng)無(wú)須游離基的參與并且沒(méi)有手性中心的產(chǎn)生。
