研究內(nèi)容
2.表觀遺傳。有些環(huán)境刺激（如飲食、毒素、應激、情緒）雖不誘發(fā)基因突變，卻可持久地改變基因功能。這些改變有時可以遺傳給后代，以致決定我們表型的，不但有基因，還有祖輩的生活經(jīng)歷甚至喜怒哀樂。這種驚人的遺傳現(xiàn)象顛覆著經(jīng)典遺傳學，但其物質(zhì)基礎(chǔ)撲朔迷離。我們建立了最簡易的小鼠模型，來攻克這一難題。
2.遺傳學技術(shù)。工欲善其事，必先利其器。我們因此研發(fā)了多種小鼠遺傳學工具,聚焦高通量篩選。對大多數(shù)基因，尤其是近年發(fā)現(xiàn)的LncRNA，我們只知序列不知功能,“知人知面不知心”。高通量篩選是揭開基因真面目的強大手段，但篩選只能在體外進行，這就嚴重地限制了其用途并構(gòu)成了遺傳學一重大瓶頸，因為多數(shù)生理現(xiàn)象無法在體外復制。我們試圖用一種嶄新的方法（Cre + Cas 9） 來打破這一瓶頸, 并用它揭示基因尤其是LncRNA 在小鼠體內(nèi)的功能。
