主要研究興趣
1. 非編碼RNA(microRNA和LncRNA)在腫瘤發(fā)生發(fā)展過程中的作用 。1) microRNA(miRNA)是一類參與基因轉(zhuǎn)錄后調(diào)控的非編碼小分子RNA,參與人體內(nèi)包括腫瘤發(fā)生在內(nèi)的多種生物學(xué)過程。有眾多的研究證實 miRNA 在腫瘤細(xì)胞中可表現(xiàn)為抑癌基因或原癌基因作用。課題組通過miRNA分析在卵巢癌中差異表達的miRNA,研究miRNA及相應(yīng)靶基因在卵巢癌早期診斷、腫瘤發(fā)生發(fā)展過程中的作用。 。2)LncRNA(長鏈非編碼RNA) 是一類轉(zhuǎn)錄本長度超過200nt的RNA分子,它們并不編碼蛋白,起初被認(rèn)為是基因組轉(zhuǎn)錄的副產(chǎn)物,不具有生物學(xué)功能。然而,近年來的研究表明,lncRNAs參與X染色體沉默, 基因組印記以及染色質(zhì)修飾,轉(zhuǎn)錄激活,轉(zhuǎn)錄干擾,核內(nèi)運輸?shù)榷喾N重要的調(diào)控過程,并與腫瘤發(fā)生有密切關(guān)系。課題組利用高通量轉(zhuǎn)錄組方法分析與卵巢癌相關(guān)的lncRNA,利用生物信息學(xué)并結(jié)合功能研究等手段探索lncRNA的功能和作用機制。
2.天然藥物的抗腫瘤機制
藥物化療是目前治療惡性腫瘤的主要手段之一,但是化療藥物的毒性反應(yīng)及腫瘤細(xì)胞產(chǎn)生耐藥性使化療藥物的使用受到限制。近年來,人們發(fā)現(xiàn)很多天然化合物如綠茶多酚、白藜蘆醇等均表現(xiàn)出明顯的抗腫瘤作用,并且能夠逆轉(zhuǎn)腫瘤細(xì)胞對化療藥物的耐藥性。本研究方向擬從天然化合物中篩選抗腫瘤活性物質(zhì),并研究它們的抗腫瘤機制及化療增敏方面的作用。
