首次將PCNA方法應(yīng)用到浮游植物細胞周期和生長率現(xiàn)場觀測,開拓了浮游植物整體細胞免疫染色的方法。應(yīng)用浮游植物整體細胞免疫染色的方法發(fā)現(xiàn)了固氮藍藻(紅束毛藻)的固氮酶只分布在一部分(分化的)細胞內(nèi),為了解此生物種如何克服固氮與光合之間矛盾提供了細胞化學(xué)的證據(jù)。應(yīng)用分子生物學(xué)和生理學(xué)方法首次發(fā)現(xiàn)在南極積雪里有細菌生存。
首次在浮游植物里克隆了線粒體細胞色素B基因,并應(yīng)用它設(shè)計分子探針用以探測噬魚費氏藻(Pfiesteria piscicida), Karlodinium veneficum等甲藻。
首次在甲藻門內(nèi)發(fā)現(xiàn)mRNA編輯現(xiàn)象,推翻了過去認為Alveolata(包括纖毛蟲、瘧原蟲)可能不存在此現(xiàn)象的假說。同時為研究甲藻(如亞力山大藻)生產(chǎn)毒素和形成赤潮的遺傳機制提供了一條新思路。
克隆了甲藻特異的Rubisco(CO2固定酶)第三例,首次證明這個基因重復(fù)排列(tandem repeat),并且這些重復(fù)基因組一起被轉(zhuǎn)錄、翻譯。用Rubisco基因首次發(fā)現(xiàn)浮游纖毛蟲攝食大型海藻,說明傳統(tǒng)的大型海藻的顆粒食物鏈需要修訂。
首次嘗試用線粒體細胞色素B基因分析浮游植物的系統(tǒng)進化,演示了這個基因作甲藻系統(tǒng)進化分析的潛力并顯示一些重要門類的系統(tǒng)地位需要修改。
首次發(fā)現(xiàn)甲藻spliced leader RNA trans-splicing, 為甲藻現(xiàn)場實時基因組表達模式的研究打下基礎(chǔ)。
在國際上首次系統(tǒng)完整地分析了甲藻基因組的結(jié)構(gòu)特性,描繪了珊瑚蟲和蟲黃藻共生過程中相互作用的分子機制,為今后甲藻基因組學(xué)和珊瑚-蟲黃藻共生生態(tài)系統(tǒng)的深入研究奠定了堅實的分子生物學(xué)基礎(chǔ)。這一研究成果不但填補了國內(nèi)空白,且處于國際領(lǐng)先水平。
