    病原細(xì)菌/人體微生物組與宿主互作-以效應(yīng)蛋白為中心

    微生物與其宿主之間的相互作用是生物物種間最復(fù)雜的相互作用之一。一方面,在與人類或者其它動植物宿主億萬年共同進(jìn)化的過程中,共棲微生物(Commensal)成功適應(yīng)了宿主環(huán)境,并對宿主的發(fā)育、免疫及神經(jīng)系統(tǒng)都產(chǎn)生重要的影響;另一方面,病原菌(Pathogens)則能突破宿主的免疫屏障,實(shí)現(xiàn)感染。雖然抗生素和疫苗的發(fā)明使我們能對付大多數(shù)的病原細(xì)菌(Pathogenic bacteria),但是,病原細(xì)菌也逐漸發(fā)展出了抵抗抗生素的策略,出現(xiàn)了超級抗性菌,使病原細(xì)菌特別是耐藥性細(xì)菌又一次成為人類的重大威脅;同時,病原細(xì)菌還發(fā)展出了精巧復(fù)雜的毒力系統(tǒng),以適應(yīng)宿主體內(nèi)環(huán)境、躲避或克服宿主的免疫體系,導(dǎo)致各種感染癥甚至癌癥等,其中最重要的毒力系統(tǒng)就是細(xì)菌的分泌系統(tǒng)(Secretion System)。通過相當(dāng)于細(xì)菌“武器”的分泌系統(tǒng),細(xì)菌可以向宿主體內(nèi)分泌數(shù)量眾多、功能各異的毒力蛋白-效應(yīng)蛋白(Effector proteins,也就是細(xì)菌的“子彈”),效應(yīng)蛋白與宿主細(xì)胞表面或宿主細(xì)胞中的各種靶點(diǎn)蛋白相互作用,改變宿主的各種信號傳導(dǎo)途徑如細(xì)胞凋亡、自噬和細(xì)胞周期等,從而適應(yīng)宿主環(huán)境,克服宿主免疫系統(tǒng)的攻擊,達(dá)到讓細(xì)菌自身在宿主內(nèi)增殖的目的。

    研究微生物和宿主相互作用的學(xué)科稱為細(xì)胞微生物學(xué)(Cellular Microbiology),細(xì)菌效應(yīng)蛋白功能的闡明是該學(xué)科研究的重點(diǎn)。(1)闡明效應(yīng)蛋白的生化功能,可以讓我們更深入理解病原細(xì)菌致病以及適應(yīng)宿主環(huán)境的分子機(jī)制;(2)以這些效應(yīng)蛋白為工具,發(fā)現(xiàn)宿主固有免疫(Innate Immunity)調(diào)控的新途徑,是免疫學(xué)的新領(lǐng)域;(3)效應(yīng)蛋白本身還可以作為抗菌藥物作用的理想靶位點(diǎn),研發(fā)針對這些效應(yīng)蛋白的活性先導(dǎo)物(相當(dāng)于鈍化或收繳細(xì)菌的“子彈”),有可能獲得避免耐藥性產(chǎn)生的新型抗生素;(4)可以運(yùn)用合成生物學(xué)(Synthetic Biology)策略在微生物細(xì)胞、癌細(xì)胞或免疫細(xì)胞中構(gòu)建效應(yīng)蛋白調(diào)控回路(Circuit) ,實(shí)現(xiàn)對疾病相關(guān)細(xì)胞活性的人工調(diào)控。

    圍繞以上方向,我們的研究興趣具體如下(詳見主頁中的“研究興趣”欄目):

    (1)效應(yīng)蛋白的生化功能及宿主免疫反應(yīng)機(jī)制以目前臨床上典型的胞內(nèi)病原細(xì)菌嗜肺軍團(tuán)菌(Legionella pneumophila)、結(jié)核分枝桿菌(Mycobacterium tuberculosis)以及重要的植物病原細(xì)菌茄科勞爾氏菌(Ralstonia solanacearum)為模型,重點(diǎn)研究其致病的關(guān)鍵效應(yīng)蛋白的表達(dá)調(diào)控及其生化功能,闡明這些蛋白與宿主靶蛋白相互作用進(jìn)而調(diào)整宿主免疫系統(tǒng)的分子機(jī)制。

    (2)靶向效應(yīng)蛋白的抗結(jié)核藥物研制結(jié)核分枝桿菌是結(jié)核病的病原,感染了我國近三分之一的人口,是國人健康的重大威脅,急需新的抗結(jié)核藥物。我們以結(jié)核分枝桿菌關(guān)鍵效應(yīng)蛋白為靶點(diǎn),建立相應(yīng)的篩選系統(tǒng),從微生物代謝產(chǎn)物中篩選效應(yīng)蛋白的抑制劑,并研究其作用機(jī)制,期望獲得新型的、能避免耐藥產(chǎn)生的新型抗結(jié)核藥物。

    (3)腸道微生物組與宿主的互作及其分子機(jī)制腸道菌群是腸道內(nèi)寄居的種類繁多的微生物的總稱。腸道微生物是代謝的重要參與者,同時代謝產(chǎn)生的脂肪酸等促進(jìn)腸道上皮細(xì)胞生長與分化。腸道菌群之間可以通過如分泌系統(tǒng)分泌的效應(yīng)蛋白和群感效應(yīng)(Quorum sensing)等維持菌群的平衡。腸道菌群失調(diào)(dysbiosis)會增加疾病的易感性,包括胃腸道疾病、代謝類疾。ㄈ绶逝、糖尿病等)、免疫類疾病、精神類疾病和癌癥等。對腸道微生物組與疾病關(guān)系的研究,將為開發(fā)基于微生物的疾病診斷、疾病干預(yù)、治療和益生菌產(chǎn)品的研發(fā)等提供基礎(chǔ)。我們綜合運(yùn)用基因組學(xué)、代謝組學(xué)和生物信息學(xué)技術(shù),研究腸道微生物組(microbiome)或腸道細(xì)菌及其效應(yīng)蛋白和代謝產(chǎn)物與炎癥和癌癥等的發(fā)生、發(fā)展的關(guān)聯(lián)性及其分子機(jī)制;同時,運(yùn)用合成生物學(xué)的相關(guān)策略,建立重要腸道菌的分子遺傳操作系統(tǒng),以期實(shí)現(xiàn)對腸道細(xì)菌功能的精準(zhǔn)調(diào)控。
