研究領(lǐng)域
離子通道的結(jié)構(gòu)和功能正常是維持生命過(guò)程的基礎(chǔ),其結(jié)構(gòu)或功能的異常會(huì)導(dǎo)致許多先天性或后天獲得性疾病,稱為離子通道病。離子通道功能的減弱或增強(qiáng)均會(huì)導(dǎo)致機(jī)體生理功能的紊亂,主要累及神經(jīng)、肌肉、心臟、腎臟等系統(tǒng)或器官。瞬時(shí)受體勢(shì)(transient receptor potential, TRP)通道是人類感覺(jué)系統(tǒng)的先驅(qū),由于可以響應(yīng)從細(xì)胞內(nèi)至細(xì)胞外的多種刺激而被稱為多覺(jué)感受器。其中的溫度敏感型TRP通道除了可被溫度激活外,還可被各種物理或化學(xué)因素激活,激活的溫敏型TRP通道不但介導(dǎo)溫度感知,而且還參與痛覺(jué)的形成和傳導(dǎo)、癢覺(jué)發(fā)生以及皮膚神經(jīng)源炎癥發(fā)生,因此,溫度敏感型TRP通道已成為治療疼痛、炎癥和瘙癢等疾病的重要新型分子靶標(biāo)。目前,我們研究小組主要聚焦于膜離子通道和神經(jīng)細(xì)胞信號(hào)傳導(dǎo)的研究,以闡述離子通道與神經(jīng)細(xì)胞信號(hào)調(diào)控的關(guān)系為主要研究目標(biāo),重點(diǎn)關(guān)注與痛覺(jué)及傷害性傳導(dǎo)密切相關(guān)的離子通道的生物學(xué)功能和作用機(jī)制。主要從以下幾個(gè)方面開(kāi)展:
1) 溫度敏感型TRP通道的門(mén)控機(jī)制與功能結(jié)構(gòu)域鑒定;
2) 重要疾病中離子通道和神經(jīng)信號(hào)調(diào)控異常的分子機(jī)制,并同時(shí)探究離子通道在生理與病理中的作用;
3) 為相關(guān)離子通道類疾病的藥物篩選和治療提供潛在的作用靶標(biāo),進(jìn)而指導(dǎo)新藥發(fā)現(xiàn)。
