通過進(jìn)化tRNA和tRNA synthetase, 我擴(kuò)展了基因密碼, 使得在活細(xì)胞中的TAG停止密碼子可以編碼熒光氨基酸,有化學(xué)反應(yīng)性的氨基酸和羥基酸. 這些技術(shù)使得我們可以在活細(xì)胞中定位不能用綠色熒光蛋白定位的蛋白質(zhì); 制備脂化或糖化的蛋白質(zhì); 并且研究蛋白折疊和酶學(xué)。 同時(shí)通過從頭設(shè)計(jì)的方法, 我也設(shè)計(jì)了含有heme的蛋白質(zhì), 并且用NMR的方法測定了結(jié)構(gòu)。
