人物簡介
蔡時(shí)青博士1997年畢業(yè)于中國農(nóng)業(yè)大學(xué)生物學(xué)院,獲理學(xué)學(xué)士學(xué)位。1997年至2002年就讀于中科院上海植物生理生態(tài)研究所,獲博士學(xué)位。2004至2009年在美國新澤西醫(yī)科牙科大學(xué)做博士后工作。2009年回國擔(dān)任中國科學(xué)院上海生命科學(xué)研究院神經(jīng)科學(xué)研究所離子通道調(diào)控研究組組長。
研究方向
鉀離子通道是一類選擇性允許鉀離子順著電化學(xué)梯度通過細(xì)胞膜的跨膜蛋白, 它存在于幾乎所有的生物體包括細(xì)菌、植物和哺乳動物中;幾乎所有活體細(xì)胞,包括肌細(xì)胞,神經(jīng)元,甚至免疫細(xì)胞中都存在鉀離子通道。因此鉀離子通道在生命過程中具有非常重要的作用:它參與了神經(jīng)元和心肌動作電位的形成,腺體激素的分泌,甚至免疫細(xì)胞的免疫反應(yīng)。鉀離子通道的功能通常是動態(tài)的,它受許多因子調(diào)控。在鉀離子通道的研究中,一個(gè)巨大的挑戰(zhàn)是鉀離子通道如何參與生命的各個(gè)生理過程,比如發(fā)育、衰老、學(xué)習(xí)和記憶的形成等等;卮疬@些基本問題,一個(gè)理想的生物體系十分重要,在這個(gè)體系里,我們能夠?qū)㈦x子通道調(diào)控和動物行為的變化聯(lián)系起來。從這一點(diǎn)講線蟲提供了一個(gè)無法估量的價(jià)值:1 幾乎所有主要的鉀離子通道家族基因都在線蟲基因組中發(fā)現(xiàn), 2 線蟲生活周期短,十分有利于遺傳學(xué)操作,3 最近發(fā)展起來的電生理技術(shù)使我們能夠在體內(nèi)或體外, 甚至線蟲活體內(nèi)測定單個(gè)神經(jīng)元細(xì)胞的電生理特征。這樣線蟲給我們提供了一個(gè)簡單的體系來回答鉀離子通道領(lǐng)域的基本問題,但又不失去普遍性。
我們課題組將結(jié)合生物化學(xué)、生物物理學(xué)、遺傳學(xué)、表現(xiàn)型分析、電生理學(xué)等方法來研究鉀離子通道在生長發(fā)育、衰老等過程中的調(diào)控機(jī)制。我們將致力于研究以下幾個(gè)問題:1 鉀離子通道α亞基和β亞基之間的相互作用機(jī)制及其生理意義;2 鉀離子通道的表達(dá)和運(yùn)輸分子機(jī)制;3 鉀離子通道在肌肉細(xì)胞衰老過程中的調(diào)控及其生理作用。另外我們也將進(jìn)行有關(guān)氯離子通道調(diào)控機(jī)制方面的研究。
